Micronuclei formation in liver fibrosis samples from patients infected by hepatitis C virus Genet. Mol. Biol.

Genetic research on fibrosis outset and its progression in chronic hepatitis (CH) by hepatitis C virus (HCV) are limited. The lack of cytogenetic data led us to investigate the presence of micronuclei (MNi), as a sign of genomic damage. Hepatocytes of hepatic parenchyma from 62 cases diagnosed with CH associated with HCV and displaying different degrees of fibrosis (F1-F4) were analyzed. These data were compared to 15 cases without fibrosis (F0). Twelve healthy liver parenchyma samples were included as control. All samples were obtained from paraffin-embedded archival material. Physalis angulata L (Solanaceae) is a medicinal plant from North of Brazil, whose different extracts and infusions are commonly used in the popular medicine for the treatment of malaria, asthma, hepatitis, dermatitis and rheumatism. However, the genotoxic effects of P. angulata on human cells is not well known. The main purpose of the present study was to evaluate the in vitro genotoxic effects of aqueous extract of P. angulata using the comet assay and the micronucleus assay in human lymphocytes provided from 6 healthy donors. Cytological and cytogenic effects induced by thiabendazole on Allium cepa root meristem JBAG

The toxicity and genotoxicity of thiabendazole (TBZ) in its commercial formulation Foldan® was studied in Allium cepa meristematic cells at concentrations ranging between 10 and 250 µg / ml. Our results show that exposure to TBZ induces a significant increase in the frequency of anaphase telophase chromosomal aberrations, micronuclei and binucleated cells at all the concentrations of TBZ assayed. The frequency of anaphase telophase aberrations was observed to reach a plateau at the concentration of 50 µg / ml of TBZ, which could be probably due to TBZ mitotic toxicity manifested at concentrations of 100 and 250 µg / ml. Three biomarkers [ethoxyresorufm-o-deethylase (EROD), DNA adducts and micronuclei (MN) were studied for biomonitoring applications along French coasts. EROD was developed at an experirnental stage in two pilot sites, Seine Bay and Fos sur Mer, using the dab (Limanda limanda), dragon et (Callionymus lyra), red mullet (Mullus barbatus) and comber (Serranus hepatus) as target species. An evaluation of the genotoxicity of domestic sewage was carried out in a municipal wastewater treatment plant. The study was carried out using Allium cepa root tip cells through cytological parameters such as aberrant cells in anaphase-telophase and the mitotic index, and the fish species Oreochromis niloticus and Tilapia rendalli, through the index of micronuclei in peripheral erythrocytes. Abstract Titanium dioxide nanoparticles (TiO2NPs) are widely used and may impact the environment. Thus, this study used a high concentration of TiO2NP (1000 mg/L) to verify the defense mechanisms triggered by a plant system – an indicator of toxicity. Furthermore, this study aimed at completely characterizing TiO2NP suspensions to elucidate their toxic behavior. TiO2NPs were taken up by meristematic cells of Allium cepa, leading to slight inhibition of seed germination and root growth. However, severe cellular and DNA damages were observed in a concentration-dependent manner (10, 100, and 1000 mg/L). The high toxicity of diesel components makes oil spills a threat to the biota in coastal marine environments. The genotoxic effect of the diesel water-soluble fraction (DWSF) on Hippocampus reidi (Ginsburg, 1933) was assessed. Fish were exposed to three different DWSF dilutions for up to 96 hours, and genotoxicity was analyzed using the micronuclei test and the comet assay. The micronuclei test revealed no significant differences between any of the DWSF dilutions and the control in the 24-hours period; however, micronuclei increased in fish exposed to 1:500 and 1:100 DWSF dilutions for 96 hours.
